Design of functional markers to determine bacterial blight resistance genes in Vietnamese local rice varieties
Bacterial blight is one of the most serious threats to many rice production regions, including Vietnam. In this study, based on the sequence data of 36 local rice varieties, candidate gene xa13 was identified in eight Vietnamese local rice varieties. The nucleotide numbers of Coding DNA Sequence of this candidate gene were similar with the published sequences in Genbank. Especially, Hom rau and Chan Thom varieties have nucleotide and amino acid ingredient similar with the published xa13 LOC Os08g42350 gene. Xa13add4 marker was designed to determine xa13 candidate genes in 36 sequenced rice varieties. The xa13add4R/xa13add4F primer pair was used to amplify the estimate 97bp DNA fragment which was similar with the published resistant rice varieties and 94bp in others. This result was very important and basis for the application of molecular markers in bacterial blight resistant rice breeding program.
