NGHIÊN CỨU XÂY DỰNG QUY TRÌNH NHÂN GIỐNG IN VITRO CÂY SÂM NÚI DÀNH

Thí nghiệm nghiên cứu xây dựng quy trình nhân giống cây Sâm Núi Dành bằng phương pháp nuôi cấy mô tế bào nhằm cung cấp cây giống chất lượng, sạch bệnh và ổn định di truyền góp phần bảo tồn và phát triển loài Sâm quý này. Kết quả nghiên cứu cho thấy khử trùng mẫu bằng HgCl2 0,1% trong thời gian 5 phút cho tỷ lệ mẫu sạch bệnh đạt 95%. Mẫu sạch được nuôi cấy trên môi trường tái sinh MS bổ sung 0,2 mg/l Ki; 1 mg/l BA; 100 ml/l nước dừa, 30 g/l đường sucroza và 6,5 g/l aga cho tỷ lệ tái sinh chồi đạt 100%. Môi trường nhân nhanh MS bổ sung 3 mg/l BA, 0,2 g/l IBA, 30 g/l đường sucroza và 6,5 g/l aga cho hệ số nhân đạt 6,6 lần. Sử dụng môi trường ra rễ 1/2MS bổ sung 1 mg/l IBA và 0,4 mg/l than hoạt tính cho tỷ lệ ra rễ đạt 95%, rễ to, mập và khỏe. Cây in vitro hoàn chỉnh được đưa ra vườn ươm trên nền giá thể hữu cơ gồm: 50% bột xơ dừa và 50% phế liệu sản xuất nấm ăn là phù hợp nhất với Sâm Nam Núi Dành, tỷ lệ sống đạt 94%.

The research experiments on in vitro propagation of Southern Nui Danh ginseng by tissue culture was carried out in order to provide high quality, free-disease and genetic stable seedlings for preserving and developing this valuable ginseng. The results showed that the sample sterilized with HgCl2 0.1% in 5 minutes had 95% free-disease rate. Sterilized samples culturing on MS shoot initiation medium (containing regeneration medium 0.2 mg/l Ki, 100 ml/l coconut water, 30 g/l sucrose and 6.5 g/l agar) had regeneration rate of 100%. Rapid MS medium of proliferation shoots including 3 mg/l BA, 0.2 g/l IBA, 30 g/l sucrose and 6.5 g/l agar could give multiplication rate to 6.6 times. The ratio of rooting on 1/2MS medium supplemented with 1 mg/l IBA, 0.4 mg/l activated carbon reached 95%. The organic substrate containing 50% of coconut powder and 50% of mushroom waste was most suitable for the in vitro Nui Danh ginseng plantlets in transferring to the nursery.
