One-step RT-PCR for detection of rice yellow stunt (Rise transitory yellowing) virus in Vietnam
One-step RT-PCR was optimized for detection of Rice yellow stunt virus (RYSV) on rice and green leafhopper, the transmission vector. Three pairs of primer were designed and selected based on the genomic sequences of RYSV downloaded from GenBank and used for detection of RYSV on infected samples in Vietnam. Different parts of infected rice plant and green leafhopper were tested. All parts of infected rice plants including fresh and dried samples can be used for the detection except the dried root. The same procedure was used and succeeded in detecting the virus from single insect of GLH collected and preserved in absolute alcohol. This protocol was used routinely at Plant Protection Research Institute for detection of RYSV on rice and GLH samples collected from different provinces in Vietnam.
