Purification of Phutho taro variety by combination of conventional method and molecular markers

Abstract

Phu Tho taro variety has been mainly grown in Tan Son, Thanh Son district, Phu Tho province. Its growth duration is from 10-12 months, plant height is from 80-100cm. The tuber of Phu Tho taro is stratified with yellow productivity. We continue to observe the characteristics and yield of these taro lines in the next gerneration. Finally, 33 taro lines of Phu Tho taro variety at the first generation was selected that revealed similar characteristics with the original variety. In the 2013, the second-crop season, these 33 taro lines were cultivated on the field to observe the germination rate. 25 elite lines were selected and evaluated agronomic traits and productivity growths, constituting parameters of these 25 elite lines on the field. Finally, 14 taro lines at 2nd generation were selected to observe some tuber parameters and tuber yield/cluster. 13 lines of 2nd generation were selected with high yield. In paralell, total DNA of these 25 elite lines were extracted from their young leaf. All of these DNA samples were screened with 55 random RAPD markers. The results obtained 12 lines showing polymorphisms with 55 RAPD markers with genetic similarity coefficient varied from 0.92-1,0. Based on analysis of genetic similarity coefficient and phenotyping evaluation on the field, 12 Phu Tho taro lines were selected. These lines revealed high genetic similarity (name: 1, 5, 7, 9, 11, 13, 14, 17, 18, 23, 24 and 25). The mixture of 12 these selected lines will be used for propagation in the next crop season.
