Study on in vitro propagation of some selected Coffea canephora clones via callus and somatic embryogenesis
The research was carried out to establish a protocol of micropropagation via callus and somatic embryogenesis from cultured leaf of some selected Coffea canephora clones in WASI. The results showed that 1g embryogenic callus could produce about 80g torpedo embryos within 4-5 months. After about 5 months of germination phase, from 1g torpedo embryos we could select 160 plantlets, size ~10 mm, with one pair of true leaves and root for acclimatization in commercial coconut fibres under a greenhouse. After 2-3 months of acclimatization, there were about 140 plantlets developed at least two pairs of true leaves available for transplanting into PE bags and taken the same care as the seedlings.
