Evaluation of genetic diversity of local rice varieties collected from Lao Cai province by using DNA markers
The genetic diversity and DNA fingerprinting of 124 local rice varieties collected from three districts of Lao Cai Provinces were elucidated by using 36 simple sequence repeat (SSR) markers distributed through 12 rice chromosomes. The result of SSR fingerprinting showed that total of 235 polymorphic bands were detected at 36 SSR loci. The numbers of polymorphic alleles varied from 3 to 14 depending on each locus, yielding average of 6 alleles per locus. The DNA profiles analysis indicated three out of 36 SSR markers revealed 3 unique alleles (rare alleles) in 3 surveyed rice genotypes. The information of these markers may come in handy to distinctly identify and characterize those 3 local varieties. Cluster analysis based on UPGMA resolved the local rice varieties into two major O. sativa groups, indica and japonica, agreed with classification based on choroplast DNA analysis. This application of DNA polymorphism analysis revealed genomic relationship in Vietnamese rice germplasm, generating a database useful for cultivar identification, local germplasm conservation, and breeding programs.
